NICD is translocated to the nucleus, where it interacts with the transcriptional regulator RBP-J or CBF1 in mammals. In the absence of a signal-receptor interaction, RBP-J represses transcription; binding of NICD to RBP-J results in the transcription of target genes including Hes1.
