Notch receptor undergoes several proteolytic steps which result in the release of the soluble intracellular domain of Notch (NICD). NICD is translocated to the nucleus, where it interacts with the transcriptional regulator RBP-J or CBF1 in mammals. In the absence of a signal-receptor interaction, RBP-J represses transcription; binding of NICD to RBP-J results in the transcription of target genes including Hes1. The deletion of RBP-J results in increased differentiation of Ngn3 cells, most likely because of decreased expression of Hes1 and other Notch targets.,
