YARS (cytosolic tyrosyl tRNA synthetase) catalyzes the reaction of tyrosine, tRNA(Tyr), and ATP to form Tyr tRNA(Tyr), AMP, and pyrophosphate. The enzyme, a class I tRNA synthetase, is a homodimer (Yang et al. 2003). Mutations in the <I>YARS</I> gene are associated with dominant-intermediate Charcot-Marie-Tooth disease â?? a form of peripheral neuropathy characterized by axon and Schwann cell dysfunction (Jordanova et al. 2006).
