<b>General Background</b> The regulatory subunits of the PI 3-kinases influence the subcellular location, enzymatic activity, and stability of the associated catalytic subunit. The family of mammalian regulatory subunits of PI 3-kinase has been slowly expanding, and currently includes six members. Three of these bind to class IA catalytic subunits (known as |FRAME: HS04527-MONOMER "p110&alpha;"|, |FRAME: HS00644-MONOMER "p110&beta;"| and |FRAME: ENSG00000171608-MONOMER "p110&delta;"|) and are activated by receptor tyrosine kinases. Two others bind to the class IB catalytic unit |FRAME: HS02818-MONOMER "p110&gamma;"|, and are activated by G protein-coupled receptors (GPCRs), and one binds to the class III catalytic |FRAME: HS01275-MONOMER "p100"| subunit. The first member of the group was |FRAME: ENSG00000145675-MONOMER "p85&alpha;"| (|FRAME: HS07270|), which contains an N-terminal src-homology 3 (SH3) domain and two src-homology 2 (SH2) domains. Other members were discovered based on homology. |FRAME: ENSG00000105647-MONOMER "p85&beta;"| (|FRAME: HS02779|) consists of similar structural domains to p85&alpha; but its amino acid sequence is only 62% identical. |FRAME: ENSG00000117461-MONOMER "p55&gamma;"| (|FRAME: HS04135|) is composed of a unique 30-residue amino terminus followed by a proline-rich motif and two SH2 domains with significant sequence identify to those in p85. It contains no SH3 domain. Two types of regulatory subunits interact only with the class IB |FRAME: HS02818-MONOMER "p110&gamma;"| catalytic subunit. They consist of |FRAME: HS13887-MONOMER "p101"| (encoded by |FRAME: HS13887|) and |FRAME: ENSG00000174083-MONOMER "p87"| (|FRAME: HS16294|) |CITS: [16476736]|. Both of these subunits bind to both p110&gamma; and heterodimeric GTPase &beta;&gamma; subunits and mediate activation of p110&gamma; downstream of G protein-coupled receptors. The last known regulatory subunit is |FRAME: HS03788-MONOMER "p150"|, encoded by |FRAME: HS03788|, which associates only with the class III |FRAME: HS01275-MONOMER "p100"| catalytic subunit. <b>About This Protein</b> The |FRAME: HS02779| gene has been mapped to chromosome 19q13.2-q13.4 |CITS: [1314371]|. A oncogenic fusion has been demonstrated between this gene and HUMORF8, a putative deubiquitinating enzyme |CITS: [9582025]|.
