The enzyme can accept both NADH and NADPH, but its affinity to NADPH is 15-fold higher than its affinity to NADH. In addition, its affinity to |FRAME: L-DELTA1-PYRROLINE_5-CARBOXYLATE| is 5-10 fold higher when NADPH is the cofactor than when NADH is the cofactor. Paradoxically, the reaction rate is about 10 fold faster when NADH is used. The Km values provided are for the enzyme with an NADPH cofactor. NADP+ inhibits the enzyme by competing with the nucleotide cofactor |CITS:[2722838]|.
