The percentages of positive lymphocytes stained with OKT3 (total), OKT4 (helper/inducer) or OKT8 (suppressor/cytotoxic) monoclonal antibodies have been determined in normal individuals. Under a variety of experimental conditions, the results were found to be unaffected by cryopreservation, paraformaldehyde fixation or incubation regime. In contrast, the results were affected, both quantitatively and qualitatively, by the choice of fluorescein-conjugated anti-mouse IgG as the second step reagent.
