A procedure for the purification of adenovirions is described. About 99 per cent pure adenovirions were obtained within only eight hours by use of chloroform treatment and two cycles of simultaneous sedimentation velocity and isopycnic centrifugation through a glycerol layer into a preformed potassium bromide gradient.
