The cleavage of substrate peptides by HIV-1 proteinase can be monitored by reaction of exposed amino groups with 2,4,6-trinitrobenzenesulfonic acid to yield yellow-colored products. This method provides a high-capacity assay of the enzyme which can be performed on microtiter plates.
