Autophosphorylation of RIP1 by RIP1.1) On TNF-alpha treatment, Rip1 undergoes autophosphorylation and ubiquitination, suggesting that one or both of these posttranslational modifications may be important in Ikk or Mkk-6 activation.2) Cells expressing kinase-inactive Rip1 failed to undergo TNF-alpha-induced phosphorylation, revealing that Tnfr1 activation stimulates Rip1 autophosphorylation.3) Rip1 autophosphorylation occurs within the cytoplasm and appears to precede Rip1 ubiquitination at the Tnfr1.4) Rip1 autophosphorylation is not required for its recruitment to the activated Tnfr1 or for its ubiquitination in lipid rafts.
