NCI: Encoded by human SKIV2L Gene (SKI2 Helicase Family), 1246-aa 138-kDa Helicase SKI2W is a conserved DEAD box protein yeast SKI2 homologue containing a leucine zipper domain, motifs associated with RNA helicases, and ATPase activity that may be involved in antiviral activity by blocking translation of poly(A) deficient mRNAs. Conserved motif DEAD box RNA helicases are implicated in translation initiation, nuclear and mitochondrial splicing, and ribosome and spliceosome assembly. Rapid 3-prime-to-5-prime degradation of unstable mammalian mRNAs containing 3-prime UTR AU-rich elements (AREs) is mediated by the multisubunit exosome. ARE-binding proteins interact with the exosome and recruit it to ARE-containing RNAs, promoting their degradation. Helicase SKI2W is an exosome-associated factor. (NCI)
