NCI: Growth Arrest and DNA-Damage-Inducible, Alpha, encoded by the GADD45A gene, is widely expressed in normal tissues and its expression is increased by DNA damage. The DNA damage-induced transcription of this gene is mediated by both p53-dependent and -independent mechanisms. The GADD45A protein responds to environmental stresses by mediating activation of the p38/JNK pathway via MTK1/MEKK4 kinase. This nuclear protein interacts with the proliferating cell nuclear antigen (PCNA), a component of DNA replication/repair complexes, and the cyclin dependent kinase inhibitor p21Cip1.
