NCI: TREX1 protein is phosphorylated by Ataxia Telangiectasia and Rad3 Related Protein (ATR), regulates ATR expression, and is an essential component of the DNA damage checkpoint pathway. TREX1 and ATR are mutually dependent partners in cell cycle checkpoint signaling pathways. The downstream ORF encodes isozymes that contain 3-prime-to-5-prime exonuclease activity and the excision of nucleotides from DNA 3-prime termini are required by DNA replication, repair, and recombination. They remove mismatched, modified, fragmented, and normal nucleotides to generate the appropriate 3-prime termini for subsequent steps in the DNA metabolic pathways. (NCI)
