NCI: TIAM1 appears to function in cellular signaling by modulating activation the GDP-GTP exchange activity of RHO-like GTPases that regulate cytoskeletal organization. The serine-rich TIAM1 protein contains a DH domain, a PDZ/DHR domain, and two PH domains. The DH domain is found in GDP-GTP exchanger proteins that activate RHO-like GTPases and PH domains are thought to be involved in protein-protein interactions. TIAM1 activates RAC1, CDC42, and to a lesser extent RHOA. (from OMIM 600687 and NCI)
