MSH: A transmembrane protein belonging to the tumor necrosis factor superfamily that specifically binds RECEPTOR ACTIVATOR OF NUCLEAR FACTOR-KAPPA B and OSTEOPROTEGERIN. It plays an important role in regulating OSTEOCLAST differentiation and activation.,NCI: Most highly expressed in lymph nodes and upregulated by TCR stimulation, human TNFSF11 Gene (TNF Family) encodes alternative isoforms of 35-kDa (precursor) homotrimeric cytokine Tumor Necrosis Factor Ligand Superfamily Member 11, which binds as an osteoclast activation and differentiation ligand to TNFRSF11B/OPG and to TNFRSF11A/RANK. Isoforms 1 and 3 are type II membrane proteins; isoform 2 is secreted. A soluble form of isoform 1 likely derives from proteolytic processing by ADAM17. TNFSF11 activates antiapoptotic kinase AKT/PKB through a SRC kinase/TRAF6 signaling complex and augments or regulates dendritic cell stimulation of naive T-cell proliferation. (NCI)
