Lanosterol synthase ((S)-2,3-epoxysqualene mutase) catalyzes the cyclization of (S)-2,3-oxidosqualene to form lanosterol during sterol biosynthesis. The crystal structure of this enzyme had been determined |CITS: [15525992]|. A human liver cDNA was expressed in <i>Saccharomyces cerevisiae</i> strain SMY8. It coded for a polypeptide of 732 amino acids, with a predicted molecular weight of 83 kDa. Its amino acid sequence was 83.3% identical to that of <i>Rattus norvegicus</i>. It also complemented an <i>erg7</i> yeast strain SMY8 |CITS: [7639730]|. A cDNA was also expressed in the yeast <i>Pichia pastoris</i> and the enzyme was purified. The molecular weight was 80,000 Da by SDS-PAGE; 85,690 Da predicted from sequence; 85,700 Da by mass spectral analysis; and 86 kDa by sedimentation equilibrium analysis in a analytical ultracentrifuge. It was determined to be most active as a monomer |CITS: [14766201]|.
